Department of Toxicology, Pharmacology and Immunotherapy is focused on

-chemical analysis of environmental aromatic hydrocarbons in both abiotic samples and food web, selected potential drugs and their metabolites and endogenous lipids (steroids, ceramides, phospholipids), using LC/MS/MS and GC/MS;
-studies on modes of action of polycyclic aromatic hydrocarbons, persistent organic pollutants and polar aromatic compounds (modulations of gene expression, DNA damage and post-geniotoxic responses, intracellular signaling, lipidomics, modulations of cell cycle, proliferation and apoptosis);
-development and validation of PCR and ELISA methods for detection of tissue- and species-specific genes and proteins;
-development of synthetic recombinant and genetic vaccines, immunotherapeutics, drug carriers including micro- and nanoparticules with focus on antiviral, antitumor and chemoprotective compounds.

The Department is organized in four research groups:

Xenobiotics in the food chain and lipidomics - head RNDr. Jiří Neča
neca__(at)__vri.cz
tel.: +420 5 3333 1819

The research team is responsible for trace analysis of chemicals from major classes of organic contaminants of the environment, foodstuffs and raw materials of animal origin (including polycyclic aromatic hydrocarbons, their derivatives and metabolites, dialkyl phthalates and other industrial and plant compounds and drugs; other task is chemical analysis of lipid cellular components and lipid signaling molecules (phospholipids, ceramides, fatty acids and steroids). The key methods are GC/MS, LC/MS/MS a HPLC/DAD.

Biochemical toxicology and ecotoxicology
- head RNDr. Miroslav Machala, CSc.
machala__(at)__vri.cz
tel.: +420 5 3333 1801

Focused on biochemical and cellular toxicology and ecotoxicology (studies on modes of action of aromatic environmental contaminants include genotoxicity, dioxin-like toxicity and other processes associated with tumor promotion and endocrine disruption). The endpoints are modulations of intracellular signal transduction pathways, gene expressions, effects of xenobiotics in plasmatic membranes and changes in cell cycle, proliferation and apoptosis status. Methodology RT-PCR, electrophoretic and microscopic methods, luminometry, fluorimetry, flow cytometry, HPLC and others.

Methods for determining food authenticity and food adulteration
– MVDr. Renčová Eva, Ph.D.
rencova__(at)__vri.cz
tel.: +420 5 3333 1617

This working group is dealing with food authenticity and food adulteration of various food commodities based on the immunochemical methods with the use of polyclonal or monoclonal antibodies against protein markers. Molecular biological methods based on the izolation of species specific DNA or tissue specific mRNA followed by applying of conventional PCR detecting systems or real-time PCR systems are also developed. Expanded authorization for the laboratory activities for the need of state veterinary inspection in the field of food adulteration was obtained.
